喉鳞癌组织中bak、bcl-2和bax蛋白的表达

摘要 目的探讨促凋亡基因 Bak 在喉鳞状细胞癌(简称鳞癌)中的表达及其与 Bcl-2、Bax 之间的关系。方法采用免疫组织化学染色方法检测45例喉原发性喉鳞癌(primary laryngeal squamous cell carcinoma,PLSCC)及10例正常喉黏膜(normal laryngeal tissues,NLT)石蜡标本组织中 Bak、Bcl-2和 Bax 蛋白的表达。结果在 NLT中阳性表达率分别为 Bak 100%(10/10)、Bcl-2 10%(1/10)和 Bax 70%(7/10);在 PLSCC 中的阳性表达率分别为Bak 44.4%(20/45)、Bcl-2 64.4%(29/45)、Bax 26.7%(12/45),两组间差异均有显著性(P<0.05);喉癌组织中Bak 蛋白的表达与肿瘤病理分级、是否有淋巴结转移显著相关(P<0.05);与患者的性别、吸烟情况、临床分期以及 Bcl-2、Bax 的表达无关(P>0.05)。Bcl-2、Bax 与肿瘤病理分级、是否有淋巴结转移、患者的性别、吸烟情况以及临床分期均无关(P>0.05);两者间的表达明显相关(P<0.05)。结论喉癌组织中 Bak 基因蛋白表达的失调可能与喉癌的发生、发展有关;Bak 的检测对喉鳞癌的治疗及估计预后有指导意义。 Purpose To investigate the expression of Bak protein in laryngeal squamous cell carcinoma and to elucidate the relationship between the expressions of Bak and expressions of the Bel-2 and Bax in the pathogenesis and development of the cancer.Methods The sections of paraffin embedded blocks were examined by immunohistochemistry for the expressions of Bak、Bcl-2 and Bax protein in 45 cases of primary laryngeal squamous cell carcinoma(PLSCC),and in 10 cases of normal laryngeal tissues(NLT).Results The expression rates of Bak、Bcl-2 and Bax in NLT were 100%(10/10), 10 %(1/10)and 70 %(7/10)respectively;and those in PLSCC were 44.4 %(20/45)、64.4 %(29/4)and 26.7 %(12/ 45)respectively,so there was significant difference between Bak、Bcl-2 and Bax in NLT compared with those in PLSCC. Expression of Bak protein in PLSCC was significantly related to pathological grade and lymph node metastases,but was not related to geoder、smoking、elinical stage and expressions of Bcl-2/Bax.Expression of Bel-2 and Bax were not related to all parameters mentioned above.Conclusion Bak may play an important role in the pathogenesis and development of PLSCC, and its expression may produce evidence for the therapeutics and prognosis of PLSCC.
