宁夏地区回族和汉族人群4个STR位点的遗传多态性分析 被引量:2

摘要 目的:分析宁夏地区回族和汉族人群D5S818、D7S820、D8S1179和D16S539 4个STR位点的遗传多态分布情况。方法:采用PCR扩增技术和聚丙烯酰胺凝胶电泳方法,分析144名无血缘关系的宁夏回族和104名宁夏汉族健康个体的遗传多态性。结果:4个STR多态位点在宁夏回族人群中分别观察到9、7、8、7个等位基因和27、19、32、23种基因型;杂合度分别为0.797 2、0.709 4、0.843 7、0.805 7:多态信息量分别为0.768 3、0.668 3、0.824 3、0.776 9;个体识别率分别为0.925 5、0.868 0、0.949 9、0.933 3;非父排除率分别为0.6289、0.495 5、0.707 9、0.639 2。在宁夏汉族人群中分别观察到8、7、8、7个等位基因和21、21、27、19种基因型;杂合度分别为0.772 6、0.774 5、0.834 2和0.835 5;多态信息量分别为0.738 8、0.741 5、0.834 2和0.835 5;个体识别率分别为0.906 6、0.919 0、0.938 6和0.922 9;非父排除率分别为0.588 0、0.590 5、0.691 3和0.689 7。结论:宁夏回族和汉族人群在这4个STR多态位点的等位基因分布有其自身的特征,且彼此之间存在着一定的差异。所得结果对进行宁夏回族和汉族人群群体遗传学研究有重要的意义。 Objective: To investigate the genetic polymorphism of 4 short tandem repeat(STR) loci of D5S818,D7S820,D8S1179 and D16S539 in Ningxia Hui and Han population. Method: The genetic polymorphism of 144 unrelated individuals of Hui population and 104 individuals of Han population from Ningxia was analyzed by polymerase chain reaction (PCR) amplification combined with denaturing polyacrylamide gel electrophoresis (PAGE) and silver staining. Results: 9, 7, 8 and 7 alleles, 27, 19, 32 and 23 genotypes of D5S818, D7S820, D8S1179 and D16S539 were detected in Ningxia Hui population; the heterozygosity (Het) of the 4 loci was 0.797 2, 0.709 4, 0.843 7 and 0.805 7 respectively; the polymorphism information content (PIC) was 0.768 3, 0.668 3, 0.824 3 and 0.776 9; the discrimina- tion power (DP) was 0.925 5, 0.868 0, 0.949 9 and 0.933 3; the paternity exclusion (PE) was 0.628 9, 0.4955, 0.7079 and 0.6392. And 8, 7, 8 and 7 alleles, 21, 21, 27 and 19 genotypes were detected in Ningxia Han population; the Het was 0.772 6, 0.774 5, 0.834 2 and 0.835 5; the polymorphism information content was 0.738 8, 0.741 5, 0.834 2 and 0.835 5; the discrimination power was 0.906 6, 0.919 0, 0.938 6 and 0.922 9; the paternity exclusion was 0.588 0, 0.590 5, 0.691 3 and 0.689 7. Conclusions: There are own characteristics, but significant differences in Ningxia Hui and Han population in the 4 STR loci, and their alleles distribution meets Hardy-Weinberg equilibrium. This is useful for population genetics in Ningxia Hui and Han population.
