摘要
目的:用基因芯片技术探讨肺鳞癌异常基因的表达。方法:提取肺鳞癌组织及正常肺组织的RNA,分别用Cy5-dCTP或Cy3-dCTP标记,再与4 096点基因芯片杂交,筛选肺鳞癌组织和正常肺组织表达差异的基因。结果:肺鳞癌组织与正常肺组织表达差异基因225条,其中癌组织中上调基因为116条, 下调基因109条;差异极显著性基因37条,表达上调24条,下调13条。结论:多基因参与肺鳞癌发病, 基因芯片技术是肺癌基因筛选的有效方法。
Objective: To screen genes with abnormal expression in lung squamous cell carcinoma with cDNA chip. Methods: The mRNA was extracted from cancer tissue and normal lung tissue, and was labeled by Cy5-dCTP or Cy3-dCTP as probes. Then the mixed probes were hybridized to cDNA chip containing 4 096 genes and genes with different expressions between lung squamous cell carcinoma and normal lung tissue were screened. Results: Two hundred and twenty-five genes were differentially expressed between squamous cell carcinoma and normal lung tissue. Compared with normal lung tissue, 116 genes were up-regulated and 109 genes were down-regulated in cancer tissue. Among the 37 genes with remarkable differential expression between the two tissues, 24 were up-regulated and 13 down-regulated. Conclusions: Many genes are possibly involved in the initiation and progression of human lung squamous cell carcinoma. cD-NA chip technique might be a useful method in screening lung cancer implicated genes.
