痉证方与痿证方对大鼠脊髓持续性压迫损伤局部细胞凋亡的影响 被引量:3

Effects of “Jingzheng Recipe” and “Weizheng Recipe” on Apoptosis at Local Continuous Compression Injury in Rat Spinal Cord

摘要 目的研究大鼠脊髓持续性压迫后细胞凋亡的变化,探讨痉、痿证方对大鼠脊髓持续性慢性损伤局部细胞凋亡的影响。方法大鼠颈椎前路螺钉压迫颈脊髓,螺钉留置持续压迫30d以产生慢性损伤,用原位末端标记技术的TUNEL法对模型组及用药组的细胞凋亡进行检测。结果脊髓受压后,灰、白质均有明显的神经细胞和胶质细胞凋亡,并以白质为明显,经药物干预后,阳性染色的神经细胞和胶质细胞减少,染色变浅。与假手术组相比,各模型组细胞凋亡数明显增高,不同压迫程度组之间有显著性差异。治疗组与模型组以及治疗组之间相比,具有非常显著性差异;痿证方组作用强于痉证方组。结论脊髓持续性压迫损伤后,受压区神经细胞和胶质细胞都有明显的凋亡;痉证方、痿证方对脊髓慢性损伤后脊髓组织神经细胞和胶质细胞的凋亡有抑制作用,痿证方强于痉证方。 Objective To study the alternation of apoptosis in rat spinal cord after continuous compression injury,and to find out the effects of “Jingzheng Recipe” and “Weizheng Recipe” on GFAP at local chronic spinal cord injury.Methods Screw was used to dill into spinal cord through anterior vertebral body.After operation the screw was remained for 30 days,and then the method of in situ end labeling TUNEL was used to check the changes of apoptosis in compressed foci.Results After spinal cord compression,the apoptosis of neurocyte and glial cell in both grey and white matter was significant,especially in white matter.After administration of “Jingzheng Recipe” and “Weizheng Recipe”,the amount of positive staining neurocyte and glial cell decreased and its staining was weakened.Compared with sham group,the apoptosis number of each group increased significantly( P <0.01).And there was a significant difference between groups of different compression degrees( P <0.05).There were also significant differences between treatment and model groups and among treatment groups ( P <0.01) .And the “Weizheng Recipe” had better effects than “Jingzheng Recipe”.Conclusions After continuous compression injury of spinal cord,the apoptosis of neurocyte and glial cell in compression location is significant.“Jingzheng Recipe” and “Weizheng Recipe” have good effects in inhibiting the apoptosis of neurocyte and glial,the latter Recipe with better effects.
