多发性骨髓瘤骨髓间充质干细胞多种细胞因子的异常表达 被引量:10

Abnormal expressions of multiple cytokines of the bone marrow mesenchymal stem cell of multiple myeloma patients

摘要 目的:了解多发性骨髓瘤(MM)病人骨髓间充质干细胞(BM鄄MSCs)的改变,进一步探讨可能存在的与MM发生﹑发展相关的MSCs异常。方法:采用半定量RT鄄PCR方法在mRNA水平上检测MM患者BM鄄MSCsIL鄄1茁,IL鄄6,SCF,TPO,G鄄CSF的表达。结果:MM组BM鄄MSCsIL鄄1茁mRNA,IL鄄6mRNA,SCFmRNA,TPOmRNA与茁鄄actin的比值分别为:0.6129±0.1702,0.9133±0.1320,0.7853±0.0943,0.6830±0.2243;非MM组BM鄄MSCsIL鄄1茁mRNA,IL鄄6mRNA,SCFmRNA,TPOmRNA与茁鄄actin的比值分别为:0.1821±0.0593,0.2371±0.0491,0.3222±0.0436,0.5400±0.2511。MM组IL鄄1茁,IL鄄6,SCF表达与正常对照组和非MM组相比均具有显著性差异(P<0.01),MM组TPO的表达与正常对照组相比差异有显著性(P<0.05),与非MM组相比差异无显著性(P>0.05)。不同组别均未见G鄄CSF的表达。结论:体外培养的多发性骨髓瘤BM鄄MSCsIL鄄1茁,IL鄄6,SCF,TPO的表达存在明显异常。 Objective:To investigate the changes of bone marrow mesenchymal stem cells(BM-MSCs) in patients with multiple myeloma(MM), further to study the abnormality of BM-MSCs related to the occurrence and development of MM. Methods:Semi-quantitiative RT-PCR was used to detect the expressions of IL-1β,IL-6,SCF,TPO at mRNA level for BM-MSCs of patients with MM. Results:The ratio of IL-1β mRNA,IL-6 mRNA,SCFmRNA,TPOmRNA to β-actin of MM were 0.6129 ± 0.1702,0.9133 ± 0.1320,0.7853 ± 0.0943,0.6830 ± 0.2243 respectively; The ratio of IL-1β mRNA,IL-6 mRNA,SCFmRNA,TPOmRNA to β-actin of non-MM were 0.1821 ± 0.0593,0.2371 ± 0.0491,0.3222 ± 0.0436,0.5400 ± 0.2511 respectively. The expressions of IL-1β,IL-6,SCF of MM had significant differences when contrasted to the normal and non-MM(P < 0.01). The expressions of TPO of MM had significant difference when contrasted to the normal(P < 0.05), but had no significant difference when contrasted to the non-MM patients(P > 0.05).Neither the MM patients nor the normal people had the expression of G-CSF. Conclusion:The abnormalities of the expressions of IL-1β,IL-6,SCF,TPO of BM-MSCs of MM cultured in vitro were positive.
