摘要
目的: 观察低强度脉冲电磁场 (pulsedelectromag neticfields,PEMFs)对体外培养成骨细胞增殖、分化、体外骨形成的影响.方法: 取出生后( <24h)的SD仔鼠 2只,骨组织块培养法分离、培养成骨细胞.频率 15Hz,场强 8Gs的PEMFs作用于成骨细胞.应用MTT方法、碱性磷酸酶测定、四环素荧光标记检测成骨细胞增殖、分化、体外骨形成情况.结果:①MTT检测结果显示对照组A490nm值为(0 330±0 020),实验组A490nm值为 ( 0 379±0 038 ),两组间有显著差异 (P<0 01).②ALP检测结果显示对照组A410nm值为 ( 0 170±0 029),实验组A410nm值为 (0 206±0 018),两组间有显著差异(P<0 01 ).③钙结节数量实验组也明显多于对照组.结论: PEMFs作用促进成骨细胞增殖、分化、加速了体外骨形成.
AIM: To observe the effects of low-intensity pulsed electromagnetic fields (PEMFs) on the proliferation,differentiation and bone tissue-like formation of cultured rat osteoblasts.METHODS: Osteoblasts isolated from newly born (<24 h) SD rats were cultured by explants outgrowth technique.The osteoblasts in the PEMFs group were cultured for 30 min in the PEMFs with the intension of electromagnetic fields 8 Gs,frequency 15 Hz.MTT assay,alkaline phosphatatase-specific activity assay and tetracycline fluorescence labeling assay were used to observe the effects of PEMFs on the osteoblasts' proliferation,differentiation and bone tissue-like formation.RESULTS: ① MTT assay demonstrated that the A 490 nm value of sham group and PEMFs group was respectively (0.330±0.020) and (0.379±0.038),with obvious difference(P<0.01).② ALP assay displayed that the A 410 nm value of sham group and PEMFs group was respectively (0.170±0.029)and (0.206±0.018),with obvious difference(P<0.01).③ The calcium node in PEMFs group was far more than that in sham group.CONCLUSION: The PEMFs can accelerate the cell proliferation,upregulate the ALP level and increase the bone tissue-like formation of rat osteoblasts in vitro.
