摘要
采用RT—PCR方法对口蹄疫病毒O/NY00株基因组L片段进行了分子克隆和序列测定。结果表明:获得的O/NY00株基因组L片段长7805nt,其中包括715at的5’非编码区和6999nt的多聚蛋白编码区。将O/NY00株与其它参考毒株进行序列比较,结果显示:O/NY00株与O/SKR/2000、O/UKG/3/2001遗传关系最近,而与其它毒株遗传关系较远,属于O型口蹄疫ME-SA拓扑型PanAsia株的成员。与Cathay拓扑型的代表毒株O/TAW/97比较,两者在主要抗原位点1和位点3呈现出明显的以拓扑型为特征的氨基酸差别,提示两拓扑型毒株之间有较高的抗原差异。
The L fragment of the genome of foot-and-mouth disease virus(FMDV) O/NY00 isolate was cloned by RT-PCR,and sequenced.The results showed that L genomic fragment of O/NY00 isolate is 7 731nt in length including partial 5' UTR and complete polyprotein coding region with 715nt and 6 999nt respectively .Comparison among O/NY00 and other reference isolates indicated that O/NY00,O/SKR/2000 and O/UKG/ 3/2001 may come from the same ancestor, and O/NY00 should be classified into PanAsia strain of FMDV serotype O ME - SA topotype. Furthermore, comparison of deduced amino acid residues at the antigenic sites with O/TAW/97,a typical isolate of Cathay topotype,showed there are obviouslly topotypical amino acid differences at Site 1 and Site 3, thus, suggesting the less antigenic relationships between these two topotype isolates.
