摘要
目的 FP248基因是本实验室通过高通量基因功能筛选得到的一个与细胞生长相关的新基因,本研究旨在研究其在肝癌发生发展中的作用。方法 用Northern blot,Southern blot方法检测FP248 在肝癌及其癌旁组织中mRNA表达差异和DNA的变化。通过体外细胞转染和体内裸小鼠成瘤实验观察FP248 对肝癌细胞SMMC 7721 生长的作用,并用Atlas Hu- man cDNA Expression Array探讨FP248影响肝癌细胞SMMC 7721的作用机理。结果 FP248的mRNA在57%(4/7)的肝癌中表达高于其相应的癌旁组织,而且肝癌组织中DNA有扩增。FP248 的过表达在体内外均可明显促进SMMC- 7721 细胞的生长。Atlas结果显示FP248过量表达后可上调许多与细胞生长密切相关的基因,同时还参与调节细胞粘附分子。结论 基因FP248与肝癌的发生发展有密切关系,是一个与人肝癌发生发展相关的新基因。
Objective To investigate the possible functions of FP248 in the carcinogenesis and progression of HCC(hepatocellalar carcinoma). Methods The increased mRNA level in HCC was examined by the Northern blot, the gene amplification in HCC was also studied by Southern blot; the promoting effect on the SMMC 7721 cells was observed by the soft agar assay,MTT method and tumor formation in nude mice. Atlas was also performed to explore the mechanism of FP248 influencing the growth of SMMC 7721 cells.Results There were both the increased mRNA and gene amplification in HCC compared with the adjacent tissues; overexpression of FP248 can promote the growth of SMMC 7721 cells in vitro and in vivo . Atlas indicated that FP248 can regulate those genes related with cell growth and other genes of cell adhesion molecules.Conclusion FP248 may be related to the HCC .
