摘要
目的 探讨环氧合酶- 2(COX- 2)mRNA在非小细胞肺癌组织中表达的临床意义。方法 提取48 例非小细胞肺癌肿瘤组织及相应癌旁组织中的RNA,应用逆转录聚合酶链反应(RT -PCR)技术检测其中COX -2mRNA的表达。结果 COX -2mRNA在48例肿瘤组织中的表达率为75%,明显高于相应癌旁组织中的表达率6%;肺腺癌的COX -2 mRNA表达率为94%,高于肺鳞癌的表达率60%(P<0.05)。COX- 2 mRNA的表达率与患者性别、年龄、组织学分化程度、原发肿瘤及淋巴结转移等因素无明显相关,而与pTNM分期关系密切(P<0.05)。COX -2 mRNA的表达强度与患者性别、年龄、病理分型及组织学分化程度等无明显相关,而与原发肿瘤、淋巴结转移及pTNM分期的关系密切(P<0.05)。结论 非小细胞肺癌组织中COX -2 mRNA的表达在非小细胞肺癌的浸润、转移和发展中具有重要意义。
Objective To study the clinical significanc e of the expression of the cyclooxygenase-2(COX-2) gene in non-small cell lun g cancer(NSCLC). Methods After the RNA was extracted from the c ancerous tissues and adjacent tissues of 48 NSCLC patients,the expressions of CO X-2 mRNA were examined by reverse transcription-polymerse chain reaction(RT-P CR). Results Expressions of COX-2 mRNA were observed in 75% of cancerous tissues and 6% of adjacent tissues respectively. The positive rates o f adenocarcinoma and squamous cell carcinoma were 94% and 60%,respectively. The expression rate was significantly higher in the adenocarcinoma than in the squam ous carcinoma. Expression rate of COX-2 mRNA was not related to patients′age a nd sex, differentiation grade of tumor cells, size of primary tumor and lymph no des metastasis, but with TNM staging. The expression intensity of COX-2 mRNA wa s not related to patients′age and sex,pathologic classification and differentia tion grade of tumor cells, but with primary tumor, lymph nodes metastasis and TN M staging. Conclusion The expression of COX-2 mRNA may play a crucial role in invasion, metastasis,and development of NSCLC.
