摘要
目的探讨通过体外循环机建立一种较为简单的乳猪隔离肺缺血再灌注模型.方法 12头4~5周乳猪,随机分为隔离肺持续灌注和缺血再灌注两组.麻醉后气管插管,从胸骨正中进胸,分别插管于肺动脉和左右心耳.引入自体血于储血器,用血泵将血经肺动脉插管泵入肺动脉,建立双肺隔离灌注模型.结果实验结束时缺血再灌注组表现为肺静止顺应性降低、湿干比增加以及光镜、电镜下出现了严重的肺组织损伤.结论本实验通过血泵建立乳猪隔离肺缺血再灌注模型具有操作简便,较之小动物实验更为接近临床的特点.为深入研究肺缺血再灌注肺损伤发生机制,以及防治措施提供良好的实验方法.
OBJECTIVE To pursue a simple method to establish isolated lung ischemia reperfusion injury model on piglets. METHODS Twelve piglets of 4-5 weeks old were randomly divided into two groups, i.e., ischemia reperfusion group and continuous perfusion group. After anesthesia and tracheal intubation, the heart was exposed by median sternotomy. Pulmonary, left atrium and right atrium cannulation was made to establish the isolated lung reperfusion model. RESULTS The decrement of lung compliance and the increment of wet to dry lung weight (W/D) ratio occurred more significantly in ischemia reperfusion group. More severe lung injury in ischemia reperfusion group was detected via light and electric microscope examination. CONCLUSION Isolated lung ischemic reperfusion model could be uneventfully established in piglets. The advantages of this model is that it is much closer to clinical settings. The establishment of this animal model facilitates the research on mechanism and prevention of ischemia reperfusion lung injury during open heart surgery.
