缺氧诱导因子-1α、血管内皮生长因子与胃癌血管生成及转移生物学行为的关系 被引量:8

Effects of Hypoxia-inducible Factor 1α and Vascular Endothelial Growth Factor on Angiogenesis and Metastasis of Gastric Carcinoma

摘要 目的 :探讨胃癌中缺氧诱导因子 1α(HIF -1α)的表达及其与血管内皮生长因子 (VEGF)、微血管密度 (MVD)的相关性 ,分析与胃癌的转移等生物学行为的关系。方法 :用免疫组化二步法检测HIF-1α、VEGF的表达 ;用CD34标记血管内皮细胞并计数MVD。结果 :胃癌组织HIF-1α及VEGF与正常胃黏膜组织比有统计学意义 (P <0 . 0 5 ) ;HIF- 1α及VEGF表达有一致性 ,呈正相关 ;HIF- 1α或VEGF阳性肿瘤组织的MVD值显著高于阴性组织 ;胃癌转移者与无转移者HIF 1α及VEGF比有统计学意义 (P <0 . 0 5 )。结论 :HIF- Objective:To explore the expression of hypoxia -inducible factor 1α(HIF-1α)and vascular endothelial growth fact o r(VEGF)in gastric carcinoma and their correlation with angiogenesis and met astasis of gastric carcinoma.Methods:Expression of HIF-1α or VEGF was assesse d by immunohistochemical staining.Microvessel density(MVD)was deter- mi ned by C D34 immunostaining of gastric carcinoma.Results:There was a significant differ ence of HIF-1α and VEGF protein expression between gastric carcinoma tissues and control tissues. There was a positive correlation between HIF-1α and VEGF . MVD in HIF-1α or VEGF positive tumor tissues was significantly higher than tha t in negative tissues.The expression level of HIF-1α or VEGF in metastasis gast r ic carcinoma was significantly higher than that in no metastasis gastric carcino ma. Conclusion: HIF-1α may play an import ant role in angiogenesis and tumor progression by regulating the expression of V EGF in gastric carcinoma.
