摘要
比较GC和Ag+-HPLC对共轭亚油酸测定的异同点。将经TLC鉴定为游离脂肪酸的样品用三甲基硅重氮甲烷甲基化,用100 m×0 25 mm的GC毛细管柱和Ag+-HPLC来测定和比较共轭亚油酸(CLA)的组成。GC和HPLC这两种方法所得的结果很相近,但各有特色。GC不能将t, t-CLA异构体8t10t、9t11t 和10t12t-CLA等分离,也不能很好地分离8t10c-CLA和9c11t-CLA;Ag+-HPLC则可以将这几种GC互相不能分离的CLA异构体分离,GC还可以将8c10t、10c12t和9t11c-CLA分离。测定CLA时最好能将两种方法结合使用。测定的两种样品CLA组成有较大的差异,反映其合成工艺有较大的不同。
The difference and homology of results in determination of conjugated linoleic acid with GC and HPLC were compared. The samples identified as free fatty acids by thin layer chromatogram (TLC) were methylated with trimethylsilyldiazomethane (TMS-DZM), then determined by GC equipped with 100 m×0.25 mm capillary column,and Ag^+-HPLC.The results of determining CLA with GC and HPLC were similar to each other, but there were differences. GC couldn't separate 8t10t,9t11t and 10t12t-CLA which come out as one peak, and not well 8t10c-CLA from 9c11t-CLA,but Ag^+-HPLC could.GC could also elicit 8c10t?10c12t and 9t11c-CLA which Ag^+-HPLC could not.It is better to combine these two methods of GC and HPLC when CLA was determined. The results also illuminated that two samples of CLA were synthesized by different methods from their different composition.
