顺铂微球的制备及其评价

摘要 以降低抗癌药顺铂的毒性,增加溶解度,加强淋巴靶向性为目的,采用S/O型乳化技术,经加热固化的方法制备了直径17±5μm的顺铂白蛋白微球。确定了最佳制备工艺,并导入对肾有保护作用的亚硒酸钠。对含药微球理化参数、LD_(50)、肿瘤抑制率等进行测定并进行了肾毒性实验。 The cisplatin albumin microspheres (the diameter is 17±5μm)were prepared by adopting the s/o type emulsive technique and using the heating-solidify method in order to reduce the toxicity of cisplantin as an anticarcinogen and increase its solubility and lymphoid target directivity. Bismuth subnitrate was added into the microspheres for protecting the renal function. In the study,a best pharmaceutical technique of preparing microspheres was found and the physical and chemical parameter, LD_(50), tumor inhibitory rate were determined. The nephrotoxicity test was also done. The results indicate that the cisplatm microspheres significantly decrease the nephrotoxicit of cisplatin.
