摘要
目的 采用体外细胞培养方法 ,研究不同浓度c9,t11-共轭亚油酸 (CLA)对人胃腺癌细胞 (SGC - 790 1)的亚油酸代谢途径中限速酶 -环氧合酶 (COX)表达的影响。方法 采用逆转录聚合酶链反应 (RT -PCR)和蛋白免疫印迹 (Westernblot)方法检测不同浓度c9,t11-CLA处理后的SGC - 790 1细胞中环氧合酶 (COX) - 1、(COX) - 2mRNA和蛋白的表达。结果 在 2 0 0 ,10 0 ,5 0和 2 5 μmol/L浓度时 ,c9,t11-CLA均可下调COX - 2mRNA和蛋白的表达 ,与共轭亚油酸 (CLA)浓度呈负相关 ;上调COX - 1mRNA的表达 ,并与CLA浓度呈正相关。结论 c9。
Objective To study the effects of c9,t1-conjugated linole ic acid (c9,t11-CLA) on the critical enzyme (COX) of linoleic acid metabolism pa th of human gastric adenocarcinoma cell line (SGC-7901). Methods Expression of COX-1,COX-2 mRNA was measured by re verse transcription polymerase chain reaction (RT-PCR) and COX-2 protein was mea sured by Western blot.Results At the concentrations of 200,100,50,25?μmol/L,c 9,t11-CLA suppressed the expression of COX-2 mRNA and protein ,but induced the e xpression of COX-1 mRNA.Conclusion Anti-cancer action of c9,t11-CLA might be asso ciated with the expression of COX.
