改良免疫-PCR检测肾综合征出血热特异性抗体

Study on modified immuno-PCR method for detection of HFRS antibodies

目的 应用改良免疫 PCR方法检测肾合征出血热 (HFRS)特异性抗体。方法 在紫外线照射处理后的PCR管中进行免疫 PCR反应 ,以链霉亲和素作为连接分子连接生物素标记二抗和生物素标记DNA ,PCR扩增DNA分子 ,琼脂糖电泳及扫描判断结果。结果 改良免疫 PCR方法的阳性检出率为 78 5 % ,常规免疫 PCR法和ELISA法的阳性检出率分别为 6 2 3%和 4 7 4 %。结论 改良免疫 PCR方法对出血热特异性抗体的阳性检出率高于常规的免疫 PCR法和间接ELISA法 ,可用于肾综合出血热特异性抗体的检测。

Objective To detect HFRS antibodies by modified immuno-PCR method.Methods Immuno-PCR reaction was operated in PCR tubes,wh ich were treated with the shining of UV ray,and streptavidin was used to bridge the biotinylated antibodies and biotinylated DNA,then the DNA was amplified in t he same PCR tube.The PCR production was analyzed in agarosegel.Results The positive rate of specific antibodies by modif i ed immuno-PCR was 78.5%,that detected by routine immuno-PCR and indirect EL ISA method were 62.3% and 47.7% respectively.Conclusion The positive rate of specific antibodies by mo dified immuno-PCR was higher than that of routine immuno-PCR and indirect ELIS A methods,so modified immuno-PCR might be used to detect HFRS antibodies.
