摘要
本文报告了反相高效液相色谱荧光检测法测定血浆或肝组织中阿霉素的含量。以正定霉素为内标物,流动相用甲醇-0.01mol/L磷酸二氢铵-冰醋酸(70:30:0.5)混合液,激发波长450nm,发射波长530nrn。血浆中药物的方法回收率101.5%,相对标准偏差2.7%;肝组织中药物的方法回收率99.6%,相对标准偏差2.0%。最低检测限6ng,最低检测浓度20ng/ml。此方法灵敏度高、简单、方便。
A high performance liquid chromatography with fluorescence detector for the determination of adriamycin(ADM) in plasma and in liver tissue was reported. Daunorubicin(DM) was used as the internal standard. The mobile phase consisted of 70ml of methanol and 30ml of 0.01mol/L NH_4H_2PO_4 solution containing 0.5ml of acetic acid. Excitation and emission wavelengths were set at 450nm and 530nm. Average recovery and RSD were 101. 5%. 2. 7% in plasma and 99. 6%. 2.0% in liver. respectively. The detection limit of adriamycin is 6ng. The method is simple and highly sensitive.
