摘要
用荧光法对EAC及EAC/ADR细胞内GSH含量进行测定,发现EAC/ADR细胞内GSH含量较EAC细胞明显增高,分别为1.25±0.35及0.31士0.12μg/106细胞;但若对EAC/ADR细胞株停用ADR治疗36周后,则能使该细胞株部分恢复其敏感性,且GSH水平也随之下降。另外,长春新碱、丝裂霉素C、放线菌素D及VP-16对EAC/ADR细胞内GSH水平均无明显影响。提示:GSH水平增高,可能是多药抗药性产生的机理之一。
Determination of GSH was performed
by modified fluorometric method.Theexperiments showed that the GSH
content in resistant cell line(EAC/ADR)was 4 fold higher than thatin
sensitive cell line(EAC),being 1. 25士0.35μg/106 cells in EAC/ADR
and 0.31士0.12μg/106cells in EAC, When the drug treatment was
discontinued for 36 weeks,the sensitivity was partiallyrecovered and
the GSH level in EAC / ADR(R)cell parallelly decreased. In addition ,
vincristine,mitomycin C ,actinomycin D and VP-16 showed no effect on
the GSH content in the EAC/ADR cell.
