EFFECTS OF OXIDIZED LOW DENSITY LIPOPROTEIN ON SMOOTH MUSCLE CELLS OF CORONARY ARTERIES

摘要 ①目的观察氧化修饰型低密度脂蛋白(OX-LDL)对培养的猪冠状动脉平滑肌细胞增殖的影响.②方法将稳定生长的2~3代猪冠状动脉平滑肌细胞分别加入25、50、100 mg/L OX-LDL和体积分数为0.03的无脂血清培养液培养24 h后,相差显微镜下观察细胞形态学变化,四唑盐(MTT)比色法测定细胞增殖能力,流式细胞术测定细胞周期各期细胞数.③结果培养猪冠状动脉平滑肌细胞在OX-LDL的作用下胞体变大,由梭形变为不规则多变形,无典型的'谷'与'峰'生长.3种不同浓度OX-LDL组细胞增殖能力均较对照组增高,差异有显著性(F=39.928,q=4.301~9.578,P<0.01);3种不同浓度OX-LDL组间差异亦有显著性(q=3.389~10.418,P<0.01).3种不同浓度OX-LDL组S期细胞百分比均显著高于对照组(χ2=10.22~17.96,P<0.01).④结论OX-LDL能直接促进体外培养的猪冠状动脉平滑肌细胞的增殖. Objective To observe the effects of oxidized low density lipoprotein (OX-LDL) on cultured smooth muscle cells(SMC) of coronary arteries in pigs. Methods OX-LDL in 25, 50, and 100 mg/L was added into the normally growing SMC of the porcine coronary arteries, respectively. 3% lipoprotein-deficient serum(LPDS) was used as the control. The morphological changes of the cells, 24 hours later, were observed under a contrast phase microscope, the cell proliferation was determined by MTT assay, and the cells number in each phase of a cell cycle was determined with the flow cytometry. Results The cultured SMC of the porcine coronary arteries enlarged with the action of OX-LDL, and their shapes changed from fusiform to irregular polygon, without typical valley and peak growth. The optical density values of the OX-LDL groups were significantly higher than those in the control group (F=39.928,P<0.01). The difference among the OX-LDL groups was also significant(q=3.389- 10.418,P<0.01). The percentage of S-phase cells increased significantly in the OX-LDL groups (χ2=10.22-17.96,P<0.01). Conclusion OX-LDL stimulates the proliferation of the cultured SMC of porcine coronary arteries.
