雌孕激素对SMMC-7721人肝癌细胞α-1,6岩藻糖基转移酶的调控 被引量:1

摘要 目的 探讨雌孕激素在肝癌细胞中对于α - 1 ,6岩藻糖基转移酶表达的调控。方法 通过对SMMC- 772 1细胞分别按不同时间点和不同浓度给予雌二醇和孕酮处理 ,观察细胞生长状态和形态学的改变 ,并进行半定量逆转录 -聚合酶链式反应和小扁豆凝集素 (LCA)印迹分析以检测α - 1 ,6岩藻糖基转移酶基因 (FUT8)转录水平和底物蛋白核心岩藻糖基化水平的改变。结果 雌激素能明显上调FUT8基因的转录 ,并显著提高Mr 为1 35× 1 0 3 、1 0 0× 1 0 3 和 6 0× 1 0 3 LCA反应阳性的糖蛋白的表达水平 ,同时改善了细胞的生长状态 ;而孕激素则可以明显下调FUT8基因的转录 ,并显著降低Mr 为 1 35× 1 0 3 、1 1 5× 1 0 3 、1 0 0× 1 0 3 和 6 0× 1 0 3 的LCA反应阳性的糖蛋白的表达水平 ,同时改变了细胞的正常形态。结论 雌孕激素能通过调节α - 1 ,6岩藻糖基转移酶的表达 ,改变靶蛋白的核心岩藻糖基化修饰 ,从而参与肿瘤的发生发展过程。 Purpose To gain insight into hormonal regulation (estrogen and progesterone) of α-1,6fucosyltransferase (α-1,6FucT) expression in hepatoma cells. Methods After SMMC-7721 cells were treated with estradiol and progesterone with different time and different concentration,cell growth and morphology were observed.The expression of α-1,6FucT gene (FUT8) in SMMC-7721 cells was assayed by semiquantitative RT-PCR and core fucosylation levels on substrates were detected by lens culinaris agglutinin (LCA) blotting. Results Estrogen could upregulate the transcription of FUT8 gene apparently and elevate the expression of LCA reactive glycoproteins of 135×103,100×103 and 60×103 Mr with the promotion of cell growth.While progesterone could markedly downregulate the transcription of FUT8 gene and reduce the expression of LCA reactive glycoproteins of 135 ×103,115×103,100×103 and 60×103 Mr with the alteration of cell morphology. Conclusions Estrogen and progesterone could alter the modification of core fucosylation on targeted proteins,thus get involved in tumorigenesis and cancer development by regulating α-1,6FucT expression in hepatoma cells.
