摘要
目的 研究亚低温对惊厥性脑损伤是否具有保护作用。方法 制作毛果芸香碱惊厥持续状态动物模型 ,给予亚低温干预 ,检测海马区神经元坏死、凋亡、凋亡蛋白表达和胶质细胞增生变化情况。结果 毛果芸香碱惊厥持续状态模型鼠病理损伤特点为神经元坏死、细胞凋亡、凋亡蛋白表达增多 ,胶质细胞增生。亚低温可减少海马区细胞坏死 :CA3区坏死细胞由 (72 .1± 1 3.4 ) %降至 (1 7.0± 6 .8) %(P <0 .0 5 ) ;减少细胞凋亡 :CA1区单位面积凋亡细胞数由 9.4± 6 .2降至 1 .0± 0 .7(P <0 .0 5 ) ;减少凋亡蛋白表达 :单位面积caspase - 3阳性面积由 1 .4 9± 0 .2 5降至 0 .6 0± 0 .1 2 (P <0 .0 5 ) ;缓解胶质细胞增生 :单位面积胶质细胞数由 4 7.3± 2 .6降至 1 4 .3±4 .4 (P <0 .0 5 )。结论 亚低温对幼鼠惊厥性神经变性具有保护作用。
Purpose To assess the neuroprotective effects of mild hypothermia on seizure-induced brain damage in rats. Methods All experimental animals (18~21 days postnatal male Wistar rats,weighing 45~50 g) were provided by laboratory animal Department of Fudan University.The rats were given 150~180 mg/kg pilocarpine (ip) to set the status epilepticus (SE) model.The SE rats were put into the cold box (6~8 ℃),maintain the rats' rectal temperature to 33~35?℃ for 72 h.The pathological changes of hippocampus were observed. Results In the SE model,seizure-induced damage could be found:neuronal necrosis,apoptosis and reactive gliolsis in the hippocampus.After 72 h mild hypothermia,the percentage of neuronal necrosis in CA3 region was significantly reduced from (72.1±13.4)% to (17.0±6.8)%;apoptosis of CA1 region was significantly reduced from (9.4±6.2) to (1.0±0.7)/unit area caspase-3 positive area per unit was significantly reduced from (1.49±0.25) to (0.60±0.12) and glial cell were significantly reduced from (47.3±2.6) to (14.3±4.4)/unit area. Conclusions Mild hypothermia may have the neuroprotective effects on seizure-induced hippocampus neuronal degeneration in rats.
