摘要
目的 观察前列腺素E1(PGE1)脂微球(Lipo PGE1)对大鼠急性胰腺炎(SAP)模型全身炎症反应(SIRS)期细胞因子调控的影响并探讨其作用机制。方法 应用5 %牛磺胆酸钠制作大鼠SAP模型,分为非治疗组和治疗组,各组内再分为3和6h组,采用逆转录 聚合酶链式反应(RT PCR)方法测定胰腺组织内肿瘤坏死因子(TNF) α、白细胞介素(IL) 6、IL 10、IL 2的表达。结果 非治疗组内3组和6h组比较,TNF α、IL 6表达逐渐增强;IL 10 ,IL 2在3h有弱表达,6h表达显著增强;治疗组3、6h的TNF α、IL 6表达同非治疗组相比明显减弱;IL 10、IL 2表达在3h无显著变化,6h明显减弱。结论 在大鼠SAP模型中,Lipo PGE1可明显抑制炎性细胞因子TNF α、IL 6和抗炎性细胞因子IL 10、IL 2的表达,使两者重新趋于平衡。
Objective To observe the effect of Lipo-prostaglandins E1 (Lipo-PGE1) on cytokine expression in phase of systemic inflammatory response syndrome (SIRS) in severe acute pancreatitis (SAP) and explore its mechanism.Methods The SAP rats were randomly divided into non-treatment and treatment group,and each group included two subgroups:3 h group and 6 h group.The expression of TNF-α,IL-6,IL-10,IL-2 in pancreas was detected by the method of RT-PCR.Results The comparison between 3 h and 6 h subgroups in non-treatment group revealed the expression of TNF-α,IL-6 was increased,and IL-10 and IL-2 had a weak expression in 3 h,but elevated significantly in 6 h.The expression of TNF-α and IL-6 was inhibited in treatment group.The expression of IL-10 and IL-2 in 3 h subgroup had no significant change,but inhibited significantly in 6 h subgroup.Conclusion Lipo-PGE1 inhibited significantly the expression of inflammatory cytokines (TNF-α,IL-6) and anti- inflammatory cytokines (IL-10,IL-2).It had an effective value on the regulation of cytokine expression in phase of SIRS in SAP.
