苦参碱联合抗肿瘤药抑制K562细胞增殖的研究 被引量:17

摘要 目的 观察苦参碱 (Ma)及Ma联合长春新碱 (VCR)、阿糖胞苷 (Ara c)、三尖杉酯碱 (HRT)、阿霉素 (ADM)、柔红霉素 (DNR)对K5 62细胞增殖的影响。方法 用MTT比色法测定Ma及Ma联合常用抗肿瘤药物对K5 62细胞的抑制率。结果 Ma( 160~ 40 0 μg/ml)对K5 62细胞有明显抑制作用。Ma ( 80 μg/ml)与VCR、Ara c、HRT、ADM、DNR分别合用时抑制率较单用VCR、Ara c、HRT时的抑制率高 (P <0 . 0 5 ) ,但较单用ADM、DNR时无明显增加抑制作用。结论 Ma对K5 62细胞增殖具有抑制作用 ,呈剂量相关性。Ma能增强VCR、Ara c、HRT对K5 Objective To investigate inhibitory effects of matrine (Ma) combined with vincristine (VCR), cytosine arabinoside (Ara-c), harringtonine (HRT), adriamycin (ADM), and daunorubicin (DNR) on proliferation of K562 cells. Methods MTT colorimetric assay was used to detect the inhibition rate of Ma combined with antineoplastic on K562 cells. Results The proliferation of K562 cells was inhibited by Ma at the concentration of 160 μg/ml to 400 μg/ml. The inhibitory rates of Ma combined respectively with VCR, Ara-c, HRT, ADM, and DNR were significantly higher than those of VCR, Ara-c, or HRT alone (P<0.05), but not significantly different from those with ADM or DNR alone (P>0.05). Conclusion Ma can inhibit the proliferation of K562 cells in a dose-dependent manner. The anti-proliferation effect of VCR, Ara-c, and HRT on K562 cells could be enhanced by Ma.
