摘要 目的 测定早孕绒毛滋养细胞中leptinmRNA表达,探讨leptin在胎盘形成过程中的可能作用。方法 取妊娠6 ~8周绒毛80份,分离、培养细胞滋养细胞并经光电镜、免疫细胞化学鉴定;提取细胞总RNA,应用逆转录聚合酶链反应(RT - PCR)技术测定滋养细胞leptinmRNA表达。结果 ①经光电镜、免疫细胞化学染色鉴定,细胞角蛋白 7(CK- 7)染色强阳性, β- hCG染色个别细胞弱阳性,vimentin染色阴性,表明分离培养的细胞为滋养细胞。②滋养细胞表达leptinmRNA在235bp处 有区带。结论 培养的早孕绒毛滋养细胞表达leptinmRNA,leptin可能在胚胎着床、滋养细胞浸润过程中起调控作用。 Objective To explore the probable action of leptin in placenta by determinating the expression of leprin mRNA in trophoblasts. Methods Cytotrophoblasts were isolated from 80 cases of the chorionic villi at 6-8-week pregnancy, cultured in vitro, and observed by light and electron microscope and immunocytochemistry. Total RNA of trophoblasts was extracted. The expression of leptin mRNA in trophoblasts was determined by reverse transcription polymerase chain reaction (RT-PCR). Results Strongly positive staining of cytokeratin-7, weakly positive staining of β-hCG in some cells, and negative staining of vimentin were observed under light and electron microscope and immunocytochemistry, suggesting they were trophoblasts. Leptin mRNA was 235 bp. Conclusion Trophoblasts from early pregnancy can express leptin mRNA which may regulate cytotrophoblast invasion in the process of embryo implantation.
