湖北土家族人群HLA-A2超型等位基因与宫颈癌相关分析

摘要 目的探讨湖北土家族人群HLA A2超型各等位基因与宫颈癌的关联。方法采用PCR/SSPOP的HLA基因分型技术,利用一对引物扩增HLA A位点2、3外显子,并通过39个地高辛标记的寡核苷酸探针分别与扩增产物杂交,进行HLA A2超型各等位基因型分型,采用卡方检验,比较宫颈癌病例组和正常对照组中HLA A2超型中相应等位基因型构成比的差异。结果HLA A0202和HLA A0206在正常对照组和宫颈癌病人组的构成比有显著性差异(P<0.05)。结论HLA A0202(OR=0.36,95%CI=0.13-0.95)和HLA A0206(OR=0.21,95%CI=0.06-0.81)对于宫颈癌发生的易感性可能有保护作用,本次研究未发现其余7种等位基因与宫颈癌有关联。 Objective To explore the association between HLA-A2 supertype alleles and cervical carcinoma. Methods Ninty-two women in whom 42 women have cervical carcinoma were conducted. 2,3 extrons of HLA-A locus were amplified. By PCR/SSOP gene typing method, a single PCR amplification was in conjunction with 39 Dig-labeled oligonucleotide probes respectively. Allele-disease associations were assessed by X 2analysis. Results Associations were observed for HLA-A*0202 with odds ratio of 0.36 (95% CI, 0.13~0.95) and HLA-A*0206 with odds ratio of 0.21(95% CI, 0.06~0.81), compared with control group. Conclusion This finding suggest HLA-A*0202 and HLA-A*0206 may be sufficient to confer protection against cervical carcinoma; however, the other 7 alleles have no association with cervical carcinoma.
