摘要
目的 探讨黄连的主要有效成分小檗碱诱导人胃癌细胞凋亡与调控基因表达水平。方法以人胃腺癌细胞株SGC - 790 1为实验模型,用盐酸小檗碱大(8μg/mL)、小(4μg/mL) 2个剂量组进行干预,采用光镜、扫描电镜、流式细胞仪对凋亡细胞进行定性与定量检测,逆转录多聚酶链式反应(RT -PCR)检测胃癌细胞中P53和Bcl- 2的mRNA表达水平。结果小檗碱8μg/mL作用72h ,电镜下出现凋亡小体;4 μg/mL、8μg/mL作用72h ,流式细胞仪DNA直方图出现典型的亚二倍体峰;4 μg/mL作用72h ,Bcl- 2基因表达水平降低,野生型P53基因表达水平升高;8μg/mL作用72h ,Bcl - 2基因不表达,野生型P53基因表达水平显著升高。结论小檗碱能诱导胃癌细胞凋亡,其机理主要为下调Bcl - 2基因的表达,上调野生型P53基因表达。
Objective To study the Effect of berberine on apoptosis and P 53 ,Bcl-2 gene transcription in human gastric cancer cell SGC-7901. The apoptosis of gastric cancer cells were detected by scanning electron microscope and flow cytometry. The mRNA expression of P 53 and Bcl-2 were assayed with RT-PCR. After 72 hours exposure to berberine of 8ug/l,apoptotic bodies were detected. There were apoptotic peaks in DNA histogram in berberine of 4ug/l and 8ug/l. The Bcl-2 mRNA transcription decreased in 4ug/l and there was no Bcl-2 in 4ug/l and 8ug/l. The wild type P 53 mRNA transcription elevated in both doses. [Conclusion]Berberine can induce apoptosis in human gastric cancer cell. The 1mechanism is to reduce the transcription of Bcl-2 and increase the P 53 . And it has the potential value in treating gastric cancer.
