妊娠高血压综合征患者PBMC培养上清液中IL-4、IL-10的检测

Detection of IL-4 and IL-10 in cell culture supernatants of PBMC in pregnancy-induced-hypertension

目的探讨Th2细胞功能在妊娠高血压综合征(妊高征)中变化。方法妊高征患者43例;正常孕妇15例;正常非孕妇女15例;分离外周血单个核细胞,在植物血凝素刺激下培养72h,采用ELISA方法培养上清液中IL-4和IL-10产生水平。结果与正常未孕组相比,正常妊娠组PBMC培养上清液中IL-4水平升高差异非常显著(P<0.001);IL-10水平升高差异显著(P<0.05)。与正常妊娠组相比,妊高征患者PBMC培养上清液中IL-4水平下降差异非常显著(P<0.01);IL-10水平有下降趋势差异没有显著性;重度妊高征患者IL-10水平下降差异显著(P<0.05)。结论正常妊娠妇女Th2细胞功能增强,而妊高征患者Th2细胞功能下降,推测妊高征的发病可能与Th2细胞功能下降有关。

To study the change of Th2 immunity in PIH. 43 patients with PIH, 15 normal pregnant women and 15 healthy nonpregnant women were selected by random. Peripheral blood mononuclear cells were seperated and cultured. Th2-type cytokines IL-4 and IL-10 in the cell culture supernatants of PBMC were detected by ELISA. Compared with normal pregnant women, IL-4 level was significantly lower in P2H group (P<0.01); IL-10 level was significantly decreased in severe PIH group (P<0.05). [Conclusions] There is increased Th2 immunity in normal pregnany and decreased Th2 immunity in PIH.
