摘要 目的 :探讨多药耐药基因 (MDR)产物在肺癌组织中的表达及其相互关系。方法 :采用免疫组化 S- P方法检测 6 0例肺癌组织中 MDR基因产物 (P- gp、GST-Л和 TOPO )的表达情况。结果 :1肺癌组织中的 P- gp、GST-Л和 TOPO 阳性表达率分别为 6 3.3%、6 6 .7%和 5 0 .0 %。GST- Л和 P- gp在非小细胞肺癌 (NSCL C)中的表达率分别为 75 %和70 .8% ,在小细胞肺癌 (SCL C)中的表达率均为 14 .3% ,两者比较均有显著性差异 (P<0 .0 5 )。GST- Л和 P- gp在腺癌的表达明显高于鳞癌 (P<0 .0 1) ;TOPO 在腺癌组的表达率明显低于鳞状细胞癌组和小细胞癌组 (P<0 .0 5 )。 2两种或两种以上 MDR基因产物的共表达率为 80 % ,明显高于单独基因的表达率 11.7% (P- gp)和 13.3% (TOPO )。GST-Л表达均伴有 P- gp和 /或 TOPO 的表达 ;P- g P表达和 GST-Л表达呈明显正相关 (P<0 .0 1)。 TOPO 表达与P- gp、GST- Л表达均呈显著负相关 (P<0 .0 1)。结论 :P- gp、GST- Л和 TOPO 在肺癌组织中的表达对肿瘤的耐药起重要作用 ,单基因和多基因协同作用 ,以多基因共表达作用为主。 Objective: To explore the expression and clinical significance of P-gp,GST-Л and TOPOⅡprotein in human lung cancer. Methods:The expression of P-gp,GST-Л and TOPOⅡprotein were detected in 60 cases of lung cancer tissue with S-P immunohistochemical method.Results:The result showed that: ① the positive expression of GST-Лand P-gp separately were 75% and 70.8% in NSCLC ,which were much higher than the positive expression in SCLC(P<0.05),compared with squamous carcinoma and adenocarcinoma, the positive rate of GST-Л and P-gp in adenocarcinoma were higher(P<0.01). But the positive rate of TOPOⅡ in adenocarcinoma was significantly lower than that in squamous carcinoma and SCLC (P<0.05).②The coexpression rate of only 2 MDR gene production and only 2 or 3 MDR gene production separately were 61.1% and 80%,which exceeded apparently more than individual expression rate 11.7% (P-gp)and 10%(TOPOⅡ); P-gp significantly positively correlated with GST-Л; P-gp and GST-Л separately had obviously negative correlation with TOPOⅡ. Conclusion:The over- expression of P-gp,GST-Л and the lower -expression of TOPOⅡ in human lung cancer made it important to influence tumor resistance , signal cooperated with many genes,but the coexpression of many genes mostly.
