摘要
以枸杞愈伤组织为材料,分别接种于对照及含100,200,400 mmol/L NaCl的培养基上,培养1,3,5,7,11 d.实验结果表明,随着盐浓度的升高,枸杞愈伤组织可溶性蛋白含量降低,且随着时间的延长呈先上升后下降的趋势,即随着盐浓度的升高,培养1,3,5 d时可溶性蛋白含量上升,5 d后开始下降;培养11 d时在不同含盐培养基上加入外源ABA,愈伤组织可溶性蛋白含量升高,通过SDS PAGE电泳实验发现,当NaCl浓度为400 mmol/L时,在愈伤组织中诱导出一条40.5 kD的新蛋白.
Lycium barbarum L.leaves were studied, which were cultured under cultural medium containing 0, 100, 200, 400 mmol/L NaCl concentration, respectively. The treatment time was 1, 3, 5, 7,11d. The results indicated that the contains of soluble proteins of Lycium barbarum L.callus decreased with increasing in salt concentration, but they increased in terms of 1, 3, 5d. After 5 days, the soluble proteins content tended to decline slowly. Meanwhile, within 11 days, when exotics ABA was put under the same conditions, they expressed increasing tendency with increasing in NaCl concentration. SDS-PAGE gel electrophoresis analysis showed that 40.5kD protein produced under 400 mmol/L NaCl stress during 11 days.
