摘要
我们提出了用掺锡的三氧化二铟 (ITO)作为工作电极 ,通过硅烷化固定化技术 ,将抗大肠杆菌O15 7:H7单克隆抗体固定在ITO电极表面 ,利用电化学阻抗谱技术来构建一种新型的免疫传感器。该新型的免疫传感器的检测限为 4× 10 3 CFU/mL ,检测线性范围为 4× 10 3 - 4× 10 6CFU/mL。实验研究表明 ,该传感器具有灵敏度较高 ,检测时间短 ,操作简单等优点 ,在临床医学和环境监测中具有应用价值。
New biosensors based on electrochemical impedance spectroscopy (EIS) were developed for detection of Escherichia coli O157:H7 through the surface immobilization of monoclonal antibody onto indium tin oxide (ITO) electrode. The biosensor could detect the target bacteria with a detection limit of 4×10 3 CFU/mL. A linear response in the electron-transfer resistance for the concentration of E.coli cells was found between 4×10 3 and 4×10 6CFU/mL. The biosensor characterizes with high sensitivity, simple operation, enabling relatively rapid and reliable analysis. The result proved the prospect of EIS biosensors on application to biomedical and environmental detection of E.coli O157:H7.
