摘要
香草醛-盐酸法、香草醛-硫酸法、正丁醇-盐酸法是测定原花青素含量的许多方法中的3种,用这3种测定方法对葡萄籽提取物中原花青素的含量进行测定,并对其实验原理、标准物的选择、标准曲线等进行讨论,然后用这3种方法测定葡萄籽提取物样品的纯度.试样在(20±1)℃条件下反应15 h,于500 nm波长下测量吸光度,线性范围为10~150 μg/mL.再用这3种标准曲线计算纯度和偏差,最后确定采用香草醛-盐酸法测定葡萄籽提取物中原花青素的含量具有较高的精确性.
Three methods,n-butyl alcohol-hydrochloric acid,vanillin-hydrochloric acid and vanillin-sulfuric acid,were used to determine the proanthocyanidin from grape seed extracts. First, the paper discussed the reaction theory, the choice of standard samples and the stand curves. Then, using three methods to determine the content of proanthocyanidin in grape seed extracts. The test wavelength was 500nm ,the linearity range was 10~150μg/mL and the reaction was finished at (20士1)℃for 15h. As a result, the vanillin-hydrochloric acid method was shown to be the best for the determination of proanthocyanidin.
