叶腋增殖途径快速繁殖欧美杂种山杨 被引量:3

摘要 以欧美杂种山杨腋芽为外植体进行组织培养,筛选出较为理想的芽分化、增殖和生根培养基,建立起有效的叶腋增殖快繁途径。选出初始培养基为WPM+BA1. 0mg·L-1;分化培养基为NT+BA1. 0mg·L-1 +KT0. 5mg·L-1;生根培养基为1 /2WPM+NAA0. 3mg·L-1 +IAA0. 05mg·L-1。根据试验结果,采用分化培养基NT+BA1. 0mg·L-1 +KT0. 5mg·L-1及抽枝培养基WPM+BA0. 3mg·L-1 +2%蔗糖对试管苗进行交替培养,观察并统计四个继代的繁殖系数,得出每个继代芽增殖系数分别为5. 22、4. 46、4. 87、5. 23。 With the tissue culture method of axillary buds, the media of differentiation, m ultiplication and rooting were chosen for the establishment of rapid propagation by multiplication of axillary buds. The best suitable initial medium was WPM+BA 1mg·L -1 ; the best suitable differentiation medium was NT+BA1.0mg·L -1 +KT0.5mg·L -1 ; the suitable rooting medium was 1/2WPM+NAA0.3mg· L -1 +IAA0.05mg·L -1 . In addition, tissue culture seedlings were cul tured by the media of NT+BA1.0mg·L -1 +KT0.5mg·L -1 and WPM+BA0.3 mg·L -1 in turn. The propagation coefficients of four subculture experim ents were 5.22, 4.46, 4.87 and 5.23, respectively.
