摘要 研究了用快速分离柱高效液相色谱法测定生物发酵液中的氨苷霉素。生物发酵液中的氨苷霉素用ZORBAX Stable Bound (4.6×50 mm,1.8 μm) C18 快速分离柱为固定相,甲醇- 0.01 mol/L的磷酸(10:90) 为流动相分离,流速为2.0 ml/min;在该色谱条件下,氨苷霉素在1.0 min内可达到基线分离;用紫外二极管矩阵检测器检测。方法标准回收率为98%~102%,相对标准偏差为0.64%~0.87%。方法用于几种生物发酵液样品中氨苷霉素的测定,取得满意结果。 A rapid column high performance liquid chromatography (HPLC) method for the determination of milidiomycin in fermentation broth was studied. The milidiomycin was separated on a ZORBAX Stable Bound (4.6×50 mm, 1.8 μm) rapid chromatographic column with methanol–0.01 mol/L phosphoric acid (10:90) as mobile phase at a flow-rate of 2.0 ml/min, and monitored with the photodiode array detector at 254 nm. The recoveries of the milidiomycin are 98%~102%, and the relative standard deviations are 0.64%~0.87%. This method was applied to the determination of milidiomycin in fermentation broth with good results.
