摘要 目的:观察13 1碘标记的血管抑素(13 1I-Angiostatin ,13 1I -AS)对小鼠Lewis肺癌移植瘤的抑制作用及其对NF -kBP6 5和CD34表达的影响。方法:30只荷Lewis肺癌的小鼠随机分成4组,分别腹腔注射13 1I-AS(含13 1I 11.1MBq ,AS 12 .5mg/kg)、AS(12 .5mg/kg)、13 1I(11.1MBq)和生理盐水各0 .3ml,每周1次,治疗2次,观察14d内肿瘤体积变化并用免疫组化法检测各组肿瘤中CD34和NF -kBP6 5的表达。结果:13 1I-AS、13 1I、AS对小鼠Lewis肺癌移植瘤的抑瘤率分别为73.3%、2 9.0 %、4 6 .4 % (与生理盐水组比,P <0 .0 5 ) ;免疫组化法检测显示,13 1I -AS治疗组中,CD34和NF -kBP6 5的表达明显低于生理盐水组(P <0 .0 1)。结论:13 1I-AS能明显抑制小鼠Lewis肺癌移植瘤的生长,降低肿瘤中CD34和NF -kBP6 5的表达,13 1I-AS在实体瘤治疗中有潜在的应用前景。 Objective:To observe the inhibition of 131 I-AS on transplanted tumor of Lewis lung carcinoma in mice and its effects on the expression of CD34 and NF -kB P65. Methods:Thirty male C57BL/6 mice with tran splanted Lewis lung carcinoma in fore limbs were divided into 4 groups. When tum ors reached 10 mm in diameter, the 4 groups were treated with 131 I (11.1 MB q per mouse), AS (12.5 mg / kg), 131I-AS (11.1MBq per mouse, 12.5 mg / kg) , and NS as control respectively. Each dr ug was given intraperitoneally and injected for two times at an interval of 7 da ys. The size of tumors was measured during the treatment, and the expression of CD34 and NF-kB P65 was detected with immunohistochemistry. Result s:① The growth of transplanted Lewis lung carcinoma was inhibite d by 73.3%, 46.4%, and 29.0% at groups treated with 131 I-AS, AS, and 131 I, respectively. ②The expression of CD34 and NF-kB P65 in 131 I -AS treatment group dropped significantly. Conclusion: 131I-AS can inhibit the growth of transplanted tumor of Lewis lung carcinoma in mice and significantly decrease the expression of CD34 and NF-kB P65 in 131 I-AS treatment group. This suggests 131 I-AS has potential prospects of clinical application in solid tumor therapy.
