摘要
以7-氨基-3-脱乙酰氧基头孢烷酸(7-ADCA)为母核,对羟基苯甘氨酸甲酯(HPGME)为酰基供体,在磷酸缓冲液体系中,利用固定化青霉素酰化酶催化合成头孢羟氨苄(cefadroxil).通过对头孢羟氨苄酶法合成过程中的影响因素的考察,以选取较佳的合成工艺条件.实验结果表明,在反应温度为25℃,反应体系pH6.5,7-ADCA,HPGME和固定化酶用量分别为2.5%,7.5%,1.5%的反应条件下,合成反应转化率可达到80.3%.
Enzymatic synthesis of cefadroxil from 7-amino-3-deactoxy-cephalosporanic acid (7-ADCA) and D-p-hydroxylphenylglycine methylester (HPGME) using immobilized penicillin acylase in phosphate buffer was investigated. Some promising results were obtained. The rate of conversion 7-ADCA to cefadroxil could reach 80.3% under the optimized conditions as follows: the temperature and pH for enzymatic reaction was 25 ℃ and 6.5, the amounts of 7-ADCA, HPGME and immobilized penicillin acylase was 2.5%, 7.5% and 1.5%, respectively.
