摘要
从15份土样和5份水样中筛选到一株产酶能力较高的细菌,经鉴定归为假单胞菌属(Pseudomonassp.),该菌产生的壳聚糖酶为诱导酶.产酶条件优化实验结果表明,产酶培养基组成为:壳聚糖10g/L,MgSO4·7H2O0.5g/L,KH2PO42.0g/L,K2HPO41.0g/L,NaCl0.5g/L,CaCl20.2g/L;最佳培养条件为:培养温度30℃,摇床转速180r/min,培养基起始pH6.0,培养时间60h,添加0.2%的蔗糖或酵母浸膏有利于产酶,在优化的培养条件下产量可达1.35U/mL.
A bacterial stain screening extracellular chitosanase was isolated from 15 soil and 5 water samples.It was identified as Pseudomonas sp..the medium composition(g/L) of producing chitosanase were as follows: chitosan 10, MgSO_4·7H_2O (0.5,) KH_2PO_4 2.0, K_2HPO_4 1.0, NaCl 0.5, CaCl2 0.2. The optimal conditions: 30 ℃,initial pH6.0, shaking at 180 r/min for 60 hours, its capacity of producing chitosanase was improved when adding 0.2% sucrose or yeast extracts in the medium. The yield of 1.35 U/mL was obtained in optimal conditions.
