硫代反义寡核苷酸质量控制方法的研究进展 被引量:1

摘要 综述了高效液相色谱(HPLC)、毛细管电泳(CE)、质谱(MS)和核磁共振(NMR)在硫代反义寡核苷酸质量控制中的应用。硫代修饰的反义寡核苷酸可以抑制基因的表达,在病毒感染和癌症治疗方面有着重要用途。药物要用于临床,必须对活性成分和生产相关的杂质进行鉴别、检查。对于硫代寡核苷酸,由于其结构特殊,必须综合运用包括HPLC ,CE ,MS和NMR在内的多种分析手段控制其质量。HPLC和CE技术用于对杂质和活性成分进行定性、定量分析;NMR可以精确地确定硫代产物的含量;MS则能够在测定硫代寡核苷酸分子量的同时确认其序列的正确性。
