M受体在烟碱提高大鼠空间工作记忆中的作用

摘要 本实验采用水迷宫中延缓的位置非匹配作业,观察M受体功能正常(安定组、安定+烟碱组)和受阻(阿托品组、阿托品+烟碱组)时,烟碱对大鼠选择游泳时的正确率和游泳时间的影响,从而探讨M受体在烟碱提高大鼠空间工作记忆中的作用。结果发现:安定降低大鼠的选择正确率(P<0. 05),增加了选择正确时的游泳时间(P<0. 05 );安定+烟碱组大鼠在训练的最后3d,选择游泳的选择正确率明显高于安定组(P<0. 05),与对照组相比无显著性差异(P>0. 05),但其选择正确时的游泳时间较对照组增加(P<0. 05);阿托品降低了选择游泳的选择正确率(P<0. 05),烟碱治疗后无明显改善(P>0. 05);阿托品组及阿托品+烟碱组的游泳时间较对照组无明显改变(P>0. 05)。本研究结果提示,烟碱长期作用过程中的增忆作用与M受体的功能增强密切相关。 The present study aimed to investigate the roles of mAChRs in nicotine-facilitated spatial working memory. Delayed nonmatching-to-position task in the water maze was used in the experiment to determine the effect of nicotine on the correct choice number and swim time for choice swim (CS) of rats, when the function of mAChRs is normal (Diazepam group and diazepam+nicotine group) or abnormal (Atropine group and atropine+nicotine group). The results indicated that diazepam significantly impaired the choice accuracy of CS ( P <0.05), and increased the swim time for correct CS ( P <0.05); However, when nicotine and diazepam were simultaneously administrated, in the last three sessions of training, the choice accuracy of CS was similar with that in control ( P >0.05), but significantly higher than that in diazepam group ( P <0.05). Meanwhile, the swim time for correct CS was longer in nicotine+diazepam group than that in control ( P <0.05). Atropine significantly decreased the choice accuracies of CS ( P <0.05); And the choice accuracy and the swim time for correct CS were not significantly different between atropine group and nicotine + atropine group in which nicotine and atropine were given simultaneously. These results indicate that the mAChRs involved in the performance of nicotine-facilitated spatial working memory in rats.
