摘要
目的 探讨干扰素引起抑郁症是否与糖皮质激素受体基因相关 ,以及干扰素如何调节糖皮质激素受体基因。方法 分别给予 3ng mL和 3 0 0ng mL剂量的干扰素诱导Jurkat细胞 1,2 ,3d ;给予 3 0 0ng/ml的干扰素诱导Jurkat细胞 3d ,然后分为两组 ,一组继续给予干扰素诱导 3d ,另一组停用干扰素观察 3d。采用Westernblot ,RT PCR和定量实时RT PCR方法分析Jurkat细胞内糖皮质激素受体基因的蛋白质和mRNA水平。结果 糖皮质激素受体基因的蛋白质和mRNA水平随着干扰素所诱导的剂量及时间的增加而下降。同时干扰素诱导的糖皮质激素受体基因表达的下降能在干扰素撤除后得到一定程度的恢复。结论 糖皮质激素受体的下降可能是干扰素引起抑郁症的发病机制之一 。
Objective To investigate the relationship between glucocorticoid receptor and the mechanism of IFN-induced depression.Methods Jurkat cells were treated with 3 ng/mL or 300 ng/mL doses of IFN for various incubation time; Jurkat cells were treated with 300 ng/ml doses of IFN for 3 days, then were divided into two groups, one was treated with IFN for additional 3 days, the other group was not treated with IFN.The protein expression and mRNA level of glucocorticoid receptor were analyzed by Western blot, RT-PCR and quantitative real-time RT-PCR. Results With different doses and in cubation time the dose- and time-dependent decreases of protein expression and mRNA level of glucocorticoid receptor were observed. Partial recovery was observed following IFN withdrawal.Conclusion The reduction of glucocortieoid receptor may be one of the mechanism of IFN-induced depression. It may provide a useful tool for predicting the development of depression.
