The expression of p53 protein and mRNA in peripheral blood monoclear cells in systemic lupus erythematosus patients

摘要 目的探讨系统性红斑狼疮(SLE)患者外周血单个核细胞(PBMC)p53mRNA及蛋白表达水平变化,从而了解p53是否通过异常的凋亡调节参与SLE的发病。方法以SLE疾病活动指数(SLEDAI)评价疾病活动性,流式细胞术检测p53 蛋白,半定量RT PCR检测p53mRNA。结果(1)SLE患者PBMC p53蛋白表达水平显著高于正常人(P<0 05)。(2)活动期与缓解期SLE患者PBMC p53蛋白的表达无显著差异, p53 蛋白水平与SLEDAI评分无关(P>0 05)。(3)SLE患者PBMC p53mRNA表达量与阳性率与正常相比无显著性差异(P>0 05)。结论(1)p53蛋白通过增加表达量而导致异常的淋巴细胞凋亡参与SLE发病,但与疾病活动性无关。(2) SLE患者PBMCp53mRNA表达未见异常。 Objective To investigate the expression of p53 protein and mRNA in peripheral blood monoclear cells(PBMC)in patients with systemic lupus erythematosus(SLE).Methods The ~index of SLE disease activity (SLEDAI)was adopted to determine the activity of SLE.The levels of p53 protein and mRNA were measured by flow cytometry and semiquantitative RT-PCR respectively.~Results (1)The percentage of p53 PBMC and the quantity of p53 in both active and inactive SLE ~patients were significantly higher than those in the normal(P<0.05).(2)No obvious difference of the expression of p53 in PBMC between the active and inactive SLE patients was found(P>0.05).And there was no significant relationship between the level of p53 protein and SLEDAI.(3)The level of p53 mRNA in PBMC of SLE patients was not obviously different from that in the normal(P>0.05).Conclusion (1)p53 may participate in SLE pathogenesis by increasing the expression of protein in PBMC,but p53 does not significantly correlate with the activity of SLE.(2)The level of p53 mRNA in PBMC of SLE patients is normal.
