摘要 目的 分析Dispase Ⅱ酶对人角膜缘干细胞培养的影响。方法 将110 例人角膜随机分为A、B 2组,在组织块培养过程中分别用和不用Dispase Ⅱ酶处理组织块,观察并分析2种处理方法培养时的细胞生长速度及培养成功率。结果 2 组细胞生长的最早及最晚时间均为(4.00±0.45)d和(11.00±0.27)d,培养成功率分别为93.54%±3.12%和98.51%±1.44%,不使用DispaseⅡ酶处理组比使用DispaseⅡ酶处理组不但培养成功率高,而且从培养至见到细胞生长时间早,统计学上有非常显著的差异(P<0 01,P<0 001)。结论 在人角膜缘干细胞的组织块培养中,不用Dispase Ⅱ酶处理可以得到更快的细胞生长速度,更高的培养成功率。 Objective To analyze the effect of Dispase Ⅱ on human limbal stem cell culture.Methods One hundred and ten human corneas were randomly divided into two groups.One group was digested by Dispase Ⅱ before culture.The time from culture to outgrow and their growing rate were observed and recorded.Results In the two groups the times from culture to outgrow were (4.00±0.45)days and(11.00±0.27)days.The growing rate were 93.54%±3.12% and 98.51%±1.44%.The group digested by Dispase Ⅱ had slower speed of cell outgrow and lower growth rate than the other group.There was a significant correlation between whether the tissue were digested by Dispasea Ⅱ and cell growth(P<0.01,P<0.001).Conclusion It is more beneficial to the human limbal stem cell culture if the tissue does not be digested by Dispase Ⅱ.
