血管紧张素Ⅱ在单基因遗传db/db糖尿病小鼠视网膜中的表达 被引量:2

摘要 目的 研究血管紧张素Ⅱ(angiotensinⅡ,AngⅡ)在C57BL/KsJ db/db糖尿病小鼠视网膜中的表达及其与病程的关系。方法 选用2 周龄db/db小鼠24 只,其db/+型同性同窝瘦型鼠为对照。取2 月龄、6 月龄、9 月龄、12 月龄C57BL/KsJ db/db糖尿病小鼠的眼球,制成石蜡切片,行免疫组织化学染色,进行免疫定位及用计算机图像分析仪对视网膜神经节细胞层的AngⅡ进行灰度和面密度分析。结果 在糖尿病早期db/db小鼠的视网膜中AngⅡ即有表达,并随病程的进展表达增强,AngⅡ主要表达在视网膜的神经节细胞层、内核层和视网膜血管。结论 在糖尿病早期眼内的AngⅡ即已激活,AngⅡ可能参与糖尿病视网膜病变的发生与发展。 Objective To study the expressions of angiotensinⅡ(AngⅡ)in retinas of C57BL/KsJ db/db diabetic mice,and investigate the correlation of AngⅡ and diabetic duration.Methods Twenty-four db/db mice of 2 postnatal weeks were selected while their thin db/+ litters as normal controls.At the end of 2 months,6 months,9 months,12 months,respectively,left eyeballs of the mice for each group were enucleated to make tissue sections for light study and immunohistochemistry analysis.The computer image-analysing instrument was used to analyze the expression of AngⅡ in ganglion cell layer.At the same time,AngⅡ in various cell layers of retina were observed and immunolocalized under light microscope.Results The expressions of AngⅡin retinas of C57BL/KsJ db/db diabetic mice were increased at the early diabetes mellitus,which was elevated continuely accompanied with the progression of diabetes duration.AngⅡimmunohistochemistry of retinas showed positive expressions in the ganglion cell layers,the inner nuclear layers and retinal vasculatures.Conclusion AngⅡwas activated at the on-set diabetes and possiblely participated in the occurrence and development of diabetic retinopathy.
