摘要
目的:探讨近距离小剂量放射治疗对血管纤粘连蛋白和细胞基质金属蛋白酶-2基因表达的影响及其机制。方法:以0、7、1 4、2 8Gy6 0 Co射线辐射培养的大鼠主动脉平滑肌细胞,观察其对纤粘连蛋白( FN)及细胞基质金属蛋白酶- 2 ( MMP- 2 ) m RNA表达的影响。结果:7Gy放疗组对细胞MMP- 2及FN m RNA表达与对照组比较无显著性差异,而1 4Gy、2 8Gy放疗组FN m RNA分别为72 .0 5±5 .82和49.77±5 .62 ,较对照组( 95 .1 7±6.41 )比较有显著性差异( P<0 .0 5 )。MMP- 2 m RNA在1 4Gy、2 8Gy放疗组分别为64.2 5±3.81和42 .1 7±3.1 6,较对照组( 82 .5 2±6.5 0 )比较也有显著性差异( P<0 .0 5 )。结论:提示1 4、2 8Gy6 0 Co射线辐射可以抑制MMP-
Objective:To study the effects of brachytherapy on the gene expession of fibronectin (FN) and matrixmetalloproteinase-2 (MMP-2) in the cultured rat vascular smooth muscle cells (VSMCs) . Methods:Rat VSMCs were cultured in DMEM containing 10% FBS and were radiated by 60 Co at a dose of 0,7,14,28Gy respectively. Then mRNA level of fibronectin and matrixmetalloproteinase-2 were studied by reveries-transcriptative competitive PCR(RT PCR) . Results: Brachytherapy inhibited the expression of MMP-2 and FN . MMP-2 mRNA decreased from 82.52±6.50 to 64.25±3.81 and 42.17±3.16 in VSMC with 14,28Gy radiation respectively( P <0.05). The FN mRNA levels were decreased from 95.17±6.41 to 72.05±5.82 and 49.77 ±5.62 in VSMC with 14,28Gy radiation respectively( P <0.05). Conclusion: Brachytheapy inhabits the expression of MMP-2 and FN in VSMC cells.
