PTEN和Ki-67在上皮性卵巢癌中的表达及意义 被引量:4

摘要 目的 探讨上皮性卵巢癌中PTEN和Ki-67的蛋白表达及意义。方法 采用免疫组织化学S-P法检测了64例上皮性卵巢癌, 30例卵巢良性肿瘤和20例正常卵巢组织中PTEN和Ki-67的表达。分析PTEN和Ki-67的表达及其与各种临床病理参数、预后的关系及二者的相关性。结果 在上皮性卵巢癌组织中PTEN表达低于正常卵巢组织和卵巢良性肿瘤组织;在病理分化差的组明显低于分化较好的组;在临床Ⅲ-Ⅳ期组PTEN表达明显低于临床Ⅰ-Ⅱ期组。而Ki-67在以上各组的表达与PTEN相比恰好相反。PTEN和Ki-67的表达与年龄、组织类型均无关。在上皮性卵巢癌中,PTEN与Ki-67的表达呈负相关。患者的生存时间与PTEN表达呈正相关。结论 PTEN蛋白的缺失与卵巢癌的发生、发展有关;PTEN蛋白表达的缺失和减少导致细胞增殖的增加,预示着卵巢癌的不良预后。 Objective To investigate the expression of PTEN and Ki -67 protein in the ovarian carcinoma and their relationship.Methods The expression of PTEN and Ki-67 protein in 64 cases of epithelial ov arian cancer,30 cases of benign ovarian cystoadenoma and 20 cases of normal ovar ian tissue were detected by S-P immunohistochemical method.Results In the nomal ovarian and benign ovarian cystoadenoma tissue,the posit ive rate of PTEN protein were higher than those in the ovarian cancer tissue. PT EN and Ki-67 protein expression varied between different clinical stages , the e xpression level of stage Ⅰ-Ⅱ group was higher than that of stage Ⅲ-Ⅳ group, P<0.05 .While the positive rate of Ki-67 were oppsite to that of PTEN.There was no different expres sion between different histological types or age. In the ovarian cancer ,there w as negtive correlation between expression of PTEN and Ki-67 protein (r = -0.594) ; In unvariate analysis , the expression of PTEN protein was as sociated with survival of ovarian cancer.Conclusion PTEN protein expression in ovarian cancer is high , expression level of PTEN protein and Ki-67 are associated with histological stage , clinic stage and survival . P TEN[ has correlation with Ki-67 ,which suggests that PTEN protein may suppress the tumor through down-regulate cell proliferation.
