摘要
目的 探讨膀胱移行细胞癌中KAI1基因mRNA的表达及其与肿瘤病理分级、临床分期及淋巴转移等的关系。方法 以β-actin为内参照,采用逆转录-聚合酶链反应(RT-PCR)检测KAI1mRNA在膀胱移行细胞癌及癌旁正常组织中的表达。结果 癌旁正常组织中均有KAI1mRNA表达,水平为1. 35±0. 38,癌组织为0. 76±0. 25,部分癌组织表达缺失,前者表达水平显著高于后者(P< 0. 01)。且随着肿瘤病理分级、临床分期的增高及淋巴结转移的出现,KAI1mRNA表达水平逐渐降低,各组间差异具有显著性意义(P< 0.01)。结论 KAI1基因表达降低与膀胱移行细胞癌分化、浸润及淋巴转移有关,有可能成为评判肿瘤恶性程度、转移潜能及预后的新指标。
Objective To explore the expression and clinical sig ni ficance of KAI1 gene in human bladder transitional cell carcinoma(BTCC). Methods Reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the expression of KAI1 mRNA in BTCC tissues and normal bladd er tissues. β-actin was the internal contrast. Results The KAI1 mRNA expression level in normal bladder tissues (1.35±0.38) was sig nificantly higher than that in BTCC tissues (0.76±0.25, P<0.01) . With the increasing of pathological grades, clinical stages and the appearance of lymph node metastasis, the expression of KAI1 mRNA decreased. There was sign ificant difference between groups (P<0.01). Conclusio n The reduction of KAI1 mRNA expression is related to the differenti ation, infiltration and lymph node metastasis of BTCC. KAI1 gene may be used as a marker to evaluate the tumor malignancy, metastasis potential and prognosis.
