β型转化生长因子在复合肌肉瓣修复骨缺损组织细胞中的表达 被引量:1

摘要 [目的]了解在复合肌肉瓣修复长段骨缺损过程中,β型转化生长因子(TGFβ)在骨组织细胞中的表达强度.[方法]取新西兰大白兔,用磨钻造成左桡骨中段1/3处直径为20mm的骨缺损,用骨蜡封闭骨髓腔,取髂骨骨松质、骨髓和桡骨处的肱肌瓣联合移植于骨缺损处,作为实验组,右侧用同样方法制成同样大小的骨缺损,仅将肱肌瓣移植于骨缺损处,任自然生长,作为对照组.分别在术后3d及1,2,4,6,8,10周处死动物,取材,固定,切片进行HE染色及TGFβ单克隆抗体免疫组织化学染色,在光学显微镜下观察.[结果]在术后3d时实验组白兔骨缺损组织细胞中的TGFβ表达呈弱阳性,1周时呈阳性,2周时呈强阳性,4~6周时呈弱阳性,8~10周时呈阴性;对照组的TGFβ表达在术后2周时呈阳性,1,4周时呈弱阳性.[结论]TGFβ是骨缺损修复中的重要因子,而复合肌肉瓣修复是治疗骨缺损的有效方法. OBJECTIVE?To study the mechanism of combined use of muscle, spongy bone particle and bone marrow (compound periosteum particle) in repairing long defects by observation of expression of TGF-β.?METHODS?20?mm standard bone defect model was procedured bilaterally in the middle radcal shaft of rabbit with bone grinding machine. Shut bone marrow socket with bone wax. taking spongy bone of ilium making particle mixing with red bone marrow from ilium repair left radical bone defect as test group. The right radical defect is left and make it growth randomly as control group. Then sacrificed at 3?rd days,1?st,2?nd,3?rd,4?th,~6?th ,8?th, 10?th weeks after surgery for histological observation immunity chemical staining of TGF-β.?RESULTS?In the test group, the immuno-histochemical staining reaction of TGF-β was weaker expression at 3?rd day and 4?th,6?th weeks after operation, the staining reaction was positive at ~1?st weeks, and highly expressed at 2?nd weeks, and weaker expressed at ~4~6 weeks, and no expressed at 8~10 weeks. In the control group, the staining reaction was positive at 2?nd weeks and weaker expressed at 1?st,4?th weeks after operation.?CONCLUSION?TGF-β is an important factors in repairing the bone defect.Genarally TGF-β expression can speak that this operation way is effective, in spongy bone combined use of spongy bone and bone marrow repairing bone defect. it can reduce the time of repairing bone defect and give some theory reliance to clinic.
