摘要
目的 检测大鼠脑缺血再灌注后细胞外信号调节蛋白激酶信使RNA(ErkmRNA)的表达,探讨降钙素 基因相关肽(CGRP)和神经生长因子(NGF)对脑组织的保护作用。 方法 采用颈动脉负压分流法制作大鼠脑缺 血再灌注模型,采用原位杂交及显微图像分析方法检测海马及皮质内Erk的表达。 结果 大鼠缺血再灌注海马 及皮质内ErkmRNA较正常组增多(P<0.05),而注射CGRP或NGF后ErkmRNA明显高于缺血再灌注组(P< 0.01),两者联合应用效果更加显著(P<0.05)。 结论 CGRP及NGF可能参与缺血神经元ErkmRNA的调节,两 者对缺血神经元有协同修复作用。
Objective To determine the expression of extracellular signal regulated kinase messenger RNA(Erk mRNA)after whole cerebral ischemia and reperfusion in rats,to explore the protective effects and mechanism of calcitonin gene-related peptide(CGRP)and nerve growth factor(NGF)on brain tissue. Methods A novel model of whole cerebral ischemia and reperfusion induced by carotid artery negative pressure shunt in rats;the expression of Erk mRNA in hippocampus and cortex was detected with in situ hybridization method and microimage analysis. Results The expressions of Erk mRNA in hippocampus and cortex were increased after cerebral ischemia and reperfusion(P<0.05);compared with this,the percentage of Erk mRNA in situ hybridization positive cells in CGRP or NGF group was higher(P<0.01);the combined usage of CGRP and NGF enhanced such effects(P<0.05).Conclusion The results indicate that CGRP and NGF take part in the regulation of expression of Erk mRNA in ischemic neurons,and they may cooperate with each other.
