摘要 目的:比较两种不同来源的CK细胞的体外扩增速度、杀伤活性和机制. 方法:脐血和正常人外周血各16例,经单个核细胞,以抗CD3mAb、重组人白细胞介素2和干扰素γ为诱导剂制备CIK细胞,直接活细胞计数法比较二者的细胞动力学效应,MTT法比较二者对胃癌细胞株BGC823的杀伤活性,透射电镜下观察肿瘤细胞的变化.结果:脐血CIK细胞的扩增速度、杀伤活性均显著优于外周血CIK细胞,CMNCCIKvsMNCCIK,P<0. 05;脐血CIK细胞以诱导肿瘤细胞坏死为主,外周血CIK细胞以诱导肿瘤细胞凋亡为主,二者的差异有统计学意义. 结论:脐血CIK细胞体外扩增快,杀伤活性强,对肿瘤细胞的作用优于外周血CIK细胞,不同来源的CIK细胞杀伤肿瘤细胞的机制不同. AIM: To compare the proliferation, cytotoxicity and the mechanism of CIK cells from two different tissues. METHODS: The peripheral blood and umbilical cord blood were harvested from 16 subjects. The lymphocytes were isolated by density gradient centrifugation and suspended in medium with CD3 mAb, rIL-2 and IFN- γ as inducing agents to prepare CIK cells. The proliferation of the cells were compared by cell counting and the cytotoxic activities to gastric carcinoma line BGC823 were determined by MTT. The changes of tumor cells after induction with CIK cells were observed with transmission electron microscope. RESULTS: The proliferation speed and the anti-tumor activity of CIK cells originated from umbilical cord blood were significantly higher than those from peripheral blood (CMNC CIK vs MNC CIK, P <0.05). CIK cells from umbilical cord blood predominantly induced the necrosis of the tumor cells while those from peripheral blood mainly induced the apoptosis of the tumor cells. The two had significant difference. CONCLUSION: Compared with the peripheral blood CIK cells, umbilical cord blood CIK cells have the advantages of rapid proliferation speed and powerful cytotoxicity. The mechanism of cytotoxicity is different in peripheral blood CIK cells and in umbilical cord blood CIK cells.
