摘要
目的 探讨单纯疱疹病毒胸腺激酶(HSV- TK)基因联合羟基无环鸟苷(GCV)对体内膀胱癌的远距离旁观者效应的效果。方法 将转染HSV -TK基因的膀胱癌细胞和未转染HSV TK的膀胱癌细胞分别种植于小鼠双侧背部;给予GCV治疗,观察肿瘤的大小变化,小鼠存活天数改变,及膀胱癌细胞病理学改变。结果 在HSV- TK/GCV系统作用下,转染HSV- TK的TK +肿瘤大小明显小于对照组,肿瘤生长受到抑制;但对侧未转染HSV -TK基因的TK 肿瘤产生远距离旁观者效应不明显。结论 HSV- TK/GCV系统对体内其他部位未转染HSV- TK基因膀胱癌细胞产生远距离旁观者效应不明显。
Objective To investigate the distant bystander effect of herpes simplex virus thymidine kinase ganciclovir (HSV-TK/GCV) system on murine bladder carcinoma cell in vivo.Methods TK+ carcinoma were induced on left flank while TK-carcinoma were induced in right flank,then the GCV treatment was given to the urime and the effect on the growth of the tumor was subsequent observed.Results In In treatment group,the TK+ tumor on the flant grow slowly significant to the tumor on the cantrol group,and the survival of the mice was prolonged.Histotlgically,there were intratumoral necrosis in the (TK+) carcinoma.But the TK-tumor on the right flant not grow slowly to the tumor on the cantrol group,and the survival of the mice was not prolonged.Histotlgically,intratumoral necrosis were not obviously in the (TK-)carcinoma.Conclusion HSV-TK/GCV oncolysates can induce the kill effect to the carcinoma cell,but the distant bystander effect to the distant carcinoma cell in other organ were not obviously.
