摘要
建立了用高效液相色谱测定食用香菇中甲醛的方法。将食用香菇用水提取 ,提取液经 2 ,4 二硝基苯肼 (DNPH)衍生后 ,用高效液相色谱法测定其中的甲醛。外标法定量测定时 ,甲醛标准溶液的峰面积与样品质量浓度在 1 0 .7~ 85 6μg/L范围内呈良好的线性关系 ,线性回归系数 (r2 )为 0 .9998,方法的检出限为8.2 μg/L。所测定的 6种香菇样品中甲醛的平均加标回收率为 72 %~ 93% ;相对标准偏差为 1 %~ 8%。除了一种野生香菇样品未检出外 ,其它 5种市售香菇样品中甲醛的质量分数为 34~ 2 92mg/kg。
A high performance liquid chromatographic (HPLC) method was developed for the determination of formaldehyde in edible shiitake mushroom. Samples are leached with water and centrifugalized, then the formaldehyde in upper centrifugate is derivatized with 2,4-dinitrophenylhydrazine (DNPH) and determined by HPLC with external standard calibration method. This proposed procedure has good linear correlation between the peak areas and formaldehyde concentrations with a dynamic linear range of 10.7~856 μg/L, a correlation coefficient (r_2) of 0.9998, and a detection limit of 8.2 (μg/L.) Formaldehyde in six shiitake mushroom samples were analyzed with spiked recoveries in the range of 72%~93%, and relative standard deviations between 1%~8%. Except for the wilding one, high contents of formaldehyde (34~292 mg/kg) were determined in all the five samples collected from local markets.
