大鼠颈动脉球囊损伤后体外培养内膜增生的形态学研究 被引量:1

摘要 目的 观察大鼠颈动脉经球囊损伤后在体外培养2周后内膜是否发生增生。方法 选取10 只健康的Wistar大鼠颈动脉球囊损伤后体外培养14d,苏木素伊红染色后进行光镜、透射电镜观察以及抗α肌动蛋白免疫组织化学染色,与正常组血管培养后进行形态学比较。结果 光镜结果可见正常组动脉血管的内膜、中膜、外膜结构完整,培养14d后内膜无明显增生,而损伤组动脉血管有新内膜形成,与正常组相比差别有显著性意义(P<0.01),新内膜细胞αSM actin免疫组织化学染色结果为阳性。电镜结果可见细胞轮廓清晰,细胞核无碎裂、溶解等现象;细胞内线粒体形态、结构正常。结论 大鼠颈动脉经球囊损伤后在体外培养2周后有新内膜形成,内膜增生是由于中膜平滑肌细胞的向内膜层的迁移和增殖。 Objective To investigate neointimal of the rat carotid arteries injuried by the balloon after organ culture for two weeks.Methods Select ten health Wistar rats and cut the rat carotid arteries after injuried by the balloon;after cultured for two weeks,to observe the injuried arteries by misroscope and transmissic electron micoscopic as well as immunohistochemistry and to compared with the normal arteries.Immunostaining of the paraffin sections was performed with anti-smooth muscle α-actin antibody.Results Misroscope indicated that the stucture of arteries of normal group was intact and the intimal unchanged .But the injuried arteries developed neointimal formation . There was significant diffrence between injuried arteries and the normal group(P<0.01). Neointimal cells were postive for α-SM-actin.Transmissic electron micoscopic indicated there was no situation of karyopyknosis and karyolysis and the mitochondria was normal.Conclusion These results suggest that medial smooth muscle cell growth and migration contribute to the neointimal formation of the rat carotid arteries injuried by the balloon after organ culture for two weeks.
